Blade of the Immortal

Manji becomes immortal after dying while seeking revenge for his sister Machi's murder. The mysterious Yaobikuni appears at his moment of death and grants him "sacred bloodworms", parasitic creatures that resurrect him as an eternal immortal. This gift comes with a curse: Manji can never truly die but must carry the guilt of all the lives he has taken and will continue to take.

Earlier scenes of Manji's guilt and the weight of his past killings foreshadow the eternal burden his immortality represents.

Manji agrees to help Rin after witnessing the horrifying sight of her mother's severed head being carried by Sabato Kuroi. This brutal image triggers Manji's own memories of losing Machi and ignites his desire to protect Rin from suffering the same fate. Makie leaves the Ittō-ryū after hearing Rin's passionate plea for vengeance against those who wronged her parents. Habaki betrays Kagehisa at Mt. Takeo due to his own ambition to rise within the Ittō-ryū ranks. Seeing an opportunity to seize power, he ambushes Kagehisa during the attempt to secure a dojo master. This treachery sets in motion the events that lead to Kagehisa's downfall and ultimately leads to Habaki becoming a target for Kagehisa's own revenge.

Despite leaving the Ittō Makie's loyalty to Kagehisa remains deeply-ryū, rooted. When he is ambushed and facing death from samurai gunfire at Mt. Takao, Makie instinctively shields him with her own body, taking the bullets instead. This act demonstrates that her feelings for Kagehisa transcended her ideological differences, showing the complexity of her character.

Makie's departure was about questioning her path, not abandoning her feelings for Kagehisa.

Manji pursues Shira with single-minded determination because Shira has kidnapped Rin, threatening everything Manji has fought to protect. Manji ultimately defeats Shira at the edge of a cliff, sending him to his death.
