Santos was killed because he was carrying a briefcase containing illegal items, dildos made from plaster casts of famous men's genitals, including Senator Gary Channel. The bartender who murdered him was working for the criminals who originally owned the briefcase. Santos was clearly nervous and anxious at the bar, suggesting he knew he was in danger but couldn't escape.

Jamie and Marian booked a one-way car service to Tallahassee but were mistakenly given a vehicle that had already been reserved for another customer, the trio of criminals led by Arliss, Flint, and The Chief. The briefcase contains a collection of dildos made from plaster casts of famous men's genitals, including Senator Gary Channel. This bizarre cargo is the reason criminals were willing to kill and pursue Jamie and Marian. The items are valuable because they can be used for blackmail, particularly the one modeled after Senator Channel, whose political career would be destroyed if the public learned about it.

Senator Gary Channel apparently had his genitals cast to create one of the dildos in the collection. The film doesn't explicitly explain how his cast ended up in the briefcase, but it suggests he was one of several prominent men whose body parts were used without their consent for this illegal collection. This becomes the key leverage Jamie and Marian use against him.

At the very end, a bellhop returns a forgotten bag containing a pair of plaster casts modeled after Senator Channel's anatomy, the same dildos that caused all the chaos throughout the film.
