In the breathtaking wilderness of Alberta’s Jasper National Park, young Omega wolf Humphrey and his friends set out to enjoy a thrilling adventure by logging a board down a steep hill. Meanwhile, the determined Alpha wolf Kate hones her hunting skills with the help of her younger Omega sister, Lilly. The paths of Humphrey and Kate fatefully intersect, but their budding connection is thwarted when Kate’s father, Winston, the leader of the Western Pack, intervenes. He reminds Humphrey of the strict pack laws that forbid Alphas and Omegas from marrying and mentions that Kate is bound for Alpha school until spring arrives.
As spring breaks the chill of winter, both Kate and Humphrey experience a transformation. Humphrey and his companions watch, captivated, as Kate embarks on her first hunt, pursuing a herd of caribou. However, the excursion takes a chaotic turn when they encounter two Eastern pack wolves, sparking a dangerous stampede. Kate heroically intervenes to save her Eastern foes from a serious threat, leading to a skirmish among the Western pack wolves, which Humphrey and his friends deftly defuse.
In a strategic meeting, Winston converses with Tony, the leader of the Eastern pack, to address a dangerous food shortage. They agree that the best solution is to merge their packs and decide that Kate should marry Tony’s Alpha son, Garth, during the upcoming Moonlight Howl to solidify the union. Kate, putting the pack’s needs above her own desires, reluctantly agrees.
When the Moonlight Howl commences, Garth embarrassingly reveals his lack of howling skills, prompting a disheartened Kate to withdraw from the event. In a twist of fate, she crosses paths with Humphrey, only for both to be unexpectedly tranquilized by park rangers, who transport them to Sawtooth National Recreation Area in Idaho. Here, they meet the witty golfing goose Marcel and his duck caddie Paddy, who enlighten them about their relocation meant to bolster their species. Determined to return to Jasper Park to prevent an impending pack war, they devise a plan.
With tensions mounting, Tony warns Winston that if Kate does not return by the full moon, conflict will be unavoidable. During this uncertain time, Lilly and Garth grow closer as she shows him around the Western pack’s territory. Meanwhile, Kate and Humphrey, who are hitching a ride on an RV, face yet another obstacle when Humphrey, needing to relieve himself, accidentally gets discovered by some people after devouring a cupcake at a gas station. As they mistakenly believe he has rabies and attempt to shoot him, Kate leaps to his rescue, but their misadventures lead them into a rainstorm, causing Kate to momentarily abandon Humphrey.
While navigating through the wilderness, Kate finds herself in a precarious situation at a flooding ravine. Humphrey comes to her aid, igniting a spark of admiration in Kate for his bravery. The next day, Marcel and Paddy assist the duo in locating a train to Jasper Park. An unexpected encounter ensues when a playful grizzly bear cub catches Humphrey’s attention, leading him and the cub to an exhilarating escape from angry grizzly bears via a wild log ride down the mountain.
As tensions rise, Lilly teaches Garth the art of howling while Humphrey’s desire for Kate compels him to howl with her, despite the laws prohibiting an Alpha and Omega from sharing such an intimate bond. Their connection deepens as they share a beautiful moment under the full moon, but their growing feelings are disrupted when Tony discovers Garth and Lilly together, prompting him to declare war against the Western pack.
While the train approaches Jasper, the dire situation escalates as humankind’s impact causes a clash between the two packs. On the day of the wedding between Kate and Garth, everything takes a dramatic turn when Kate publicly professes her love for Humphrey. In response, Garth reveals his feelings for Lilly, triggering an upheaval that leads to an all-out brawl. Amidst the chaos, the sound of the stampede of caribou draws attention, forcing Winston and Tony to flee for their lives.
In a remarkable display of teamwork, Kate and Humphrey unite to save their respective packs from the stampede, but tragedy strikes when Kate is struck by a caribou. As the chaos settles, Humphrey desperately howls for Kate, awakening a powerful response from everyone around him. In a poignant reunion, Kate awakens and confesses her love for Humphrey, which matches the feelings expressed by Garth and Lilly.
Ultimately, Winston and Tony abandon the old laws prohibiting inter-pack marriages, welcoming a new era of unity through the marriage of Garth and Lilly. At the Moonlight Howl, the wolves celebrate this monumental shift in tradition, as Kate and Humphrey, along with Garth and Lilly, joyfully howl a harmonious duet, bridging the gap between Alphas and Omegas in a vibrant display of love and acceptance.
