Tom [Bill Kopp] chases Jerry from their house and across town until they reach the International Space Place, where astronauts Buzz Blister [Jess Harnell] and Biff Buzzard [Billy West] are heading to Mars. In the chaos of a speech, the duo is mistaken for aliens after Tom is splashed with green paint backstage, and the staff move to capture them. Although Jerry manages to slip away, Tom is caught and ejected from the scene.
As the staff test dehydrated food, chaos erupts when Jerry accidentally spills a cup, sending the food flying and triggering a popping explosion. The scene escalates quickly: the staff pursue Jerry, but they realize that only Tom might be able to catch him. They bring Tom back to the base and assign him a mission to eliminate Jerry, turning the usual chase into a declared search and a test of whose wits will prevail.
The chase rockets them to Mars when their path leads onto a rocket, and the two friends end up stranded on the red planet. A green female alien named Peep [Kathryn Fiore], her alien dog Ubu, and three other Martians arrive, and Jerry is taken to the Martians’ lair. There, he is briefly mistaken for the fabled “Great Gloop,” which creates a bout of confusion and danger. After the misunderstanding is cleared, Tom, Jerry, and Peep hatch a plan to hijack a flying saucer and hurry back to Earth to warn everyone about a potential Martian attack. Their quick thinking helps them reach Earth just in time to mount a defense against the looming threat.
But the Martians’ plan is far from over. A towering orange vacuum-cleaner robot known as the Invince-a-tron arrives on Earth and begins to sweep up people and places in its relentless suction. Tom, Jerry, and Peep race against the clock, using a clever ruse to get Spike—the Martian guard dog—into the Invince-a-tron’s brain with a bone, causing the machine to malfunction and ultimately destroying it.
In gratitude, Tom and Jerry are awarded a shiny Hummer by the U.S. President Jeff Bennett for saving Earth from invasion. Their victory, however, is short-lived: a repaired Invince-a-tron controlled by Spike returns for revenge, threatening to finish what it started. Peep swoops in aboard a flying saucer to rescue Jerry, but Tom is left to be pursued by the Spike-controlled Invince-a-tron. The epilogue follows Biff Buzzard and Buzz Blister cleaning up the mess as punishment for their earlier lies about Mars’ life, soon slipping into a petty argument that ends with Tom still dodging danger as the sun sets on his latest Earth-crossing adventure.
