Milady targets Constance because she serves as the Queen's lady-in-chief and is the primary messenger between Anne and Buckingham. By eliminating Constance, Milady can disrupt the Queen's communications and advance Richelieu's scheme. At the end of the film, Milady's henchmen abduct Constance, leading to D'Artagnan being knocked unconscious while trying to rescue her.

Foreshadowing: Constance's role as the Queen's confidante makes her a natural target for Richelieu's network.
