In Carpathia, a group of rogue KGB agents plots to resurrect Vlad Dracula using the Judas Chalice, intending to resurrect the old empire of evil. Their scheme sets the stage for a race to control the relics and the power they grant. The plan soon threads toward New Orleans, where the chalice is believed to be hidden.

In Bucharest, Lazlo is abducted by Kubichek's men and drawn into the chase for the Chalice. The elderly historian is coerced into helping locate the relic and is steered toward New Orleans, the key destination in the hunt.

Back at the Library, Flynn's financial and personal struggles boil over, prompting Judson, Charlene, and Excalibur to suggest a vacation. The crisis underscores Flynn's unsettled destiny and the looming fight between good and evil.

Charlene drops by with travel brochures and urges Flynn to follow his dreams. That night, Flynn's subconscious visions pull him toward New Orleans, setting the stage for the pursuit of the Chalice.

Flynn travels to New Orleans, guided by his strange dreams. In a church-turned-nightclub, he meets Simone Renoir, the woman in his dreams, who guards the first marker for locating the Chalice. After a tense escape from Kubichek's men, Flynn and Simone spend the night together.

Judson appears in the morning and explains that the Chalice is the vampire version of the Holy Grail, formed from Judas's betrayal. He teaches Flynn how staking vampires works, emphasizing aspen wood as the only effective material. Flynn absorbs the crucial knowledge to survive the hunt.

With Andrew's guidance, Flynn deciphers the markers and travels to Marie Laveau's tomb to locate the second marker. Kubichek's henchmen close in, capturing Flynn and Lazlo as the chase intensifies through the streets of New Orleans.

Flynn and Simone fight their pursuers through a carnival-laden street party and reach the wrecked pirate ship where the Chalice is hidden. The duo faces grave danger, with Simone displaying formidable powers as the danger closes in.

After a brutal confrontation, Flynn awakens in Simone's home and learns she has guarded the Chalice for two centuries. She shares a 1,000-year-old painting depicting the Library's ancient struggle, revealing the Scholar/Yahuda motif. Flynn realizes Judson may be the long-lost Yahuda from the painting.

On the bayou's wreck, Lazlo drinks from the Chalice and becomes Vlad Dracula, the creature foretold by the legends. Kubichek is transformed in turn, turning the would-be army into a new vampire force loyal to Vlad. The true scale of the threat becomes terrifyingly clear.

Flynn tracks Vlad outside and attempts to reclaim the Chalice, while Ivan drives Simone away and the hideout is blown up. An aspen stake ends Lazlo's unholy reign, and Vlad's plans begin to unravel as Kubichek meets his end in the explosion.

With Lazlo defeated, Simone asks Flynn to watch one last sunrise with her. They share a goodbye kiss as she dissolves into sparkles, choosing peace after centuries of fighting her maker. Flynn is left to carry on the fight with renewed purpose.

Flynn returns to the Library and passes the first marker to Charlene on a cord. He teases Charlene with the inscription 'follow your dreams,' confirming that her advice led him home. Judson's true identity as Yahuda is implied, and the Tree of Knowledge signature appears on the Library itself.
