Spot has impersonated a boy named Scott for the past year and attends Leonard Helperman's school. He longs to become a real human boy, which frustrates Leonard who just wants a normal life for his dog. This setup drives the central conflict of their unusual arrangement.

Mary Lou Helperman is invited to the Teacher of the Year finals in Sunny Southern Florida and travels there by the principal's RV, with the condition that no dogs are allowed inside. Leonard must bid farewell to Spot, who stays behind with a pet sitter. The trip seeds the events that bring Spot and Leonard toward Krank's lab.

While at home, Spot watches a televised talk show where Dr. Ivan Krank claims he can turn animals into human beings. The boastful promise sparks Spot's determination to meet Krank and become a boy. He resolves to chase the RV to Florida to seek him out.

Spot disguises himself as Scott and persuades Mrs. Helperman to allow him to travel with them. He joins the RV journey in hopes of reaching Krank's lab. Leonard is left to wonder about the strange new 'Scott' alongside him.

Back home, the Helpers' bird Pretty Boy and cat Mr. Jolly learn from another talk show episode that Krank cannot truly turn animals into people, only into grotesque hybrids. They decide to follow Spot to warn him. This knowledge raises the stakes for Spot's plan.

They arrive in Florida and Mary Lou heads straight to the finals, while Spot convinces Leonard to accompany him to Krank's lab. The dynamic shifts as Leonard weighs his loyalty against his fear. The stage is set for the transformation.

Krank agrees to turn Spot into a human, gleefully anticipating the respect denied him by the scientific community. The procedure is framed as a way to grant Spot a human life. The moment marks a dramatic turning point in their quest.

Spot awakens to find he is human, but dogs age faster, so he is a fully grown man rather than a boy. He retains dog instincts while now living as a man. Krank's plans to showcase the 'dog-man' become immediately apparent.

Krank intends to capture and exhibit the dog-man, but Spot and Leonard break free. Scott uses his dog senses to locate a lost dog and win a cash reward, and he and Leonard enjoy a day on the town. Their bond deepens as they navigate their new reality.

Back at the RV, they fabricate a story explaining Scott's disappearance and his new human form. Mrs. Helperman invites Scott in for coffee and starts to fall for him. Scott contemplates marrying Mary Lou so they can all stay together, but Leonard refuses.

Krank plans to turn Leonard into a dog and capture Scott to exhibit them together. Scott arrives, destroys Krank's machine, and chaos erupts; Krank is turned into a mouse and Scott returns to his canine form. The villain is defeated and balance begins to return.
