When the show boat Cotton Blossom arrives in a Mississippi town to stage a performance, a tense fistfight breaks out between the leading man, Steven Baker, and Pete, the boat’s engineer who has been making passes at Baker’s wife, the leading lady, Julie La Verne. Pete knows a dark secret about the couple: Julie is part black, and therefore their marriage is illegal in the eyes of the town. After Pete arrives with the sheriff, Baker pricks Julie’s finger and sucks a drop of her blood, a stark moment that suggests he now bears “negro blood” too. Because black and white actors cannot share the stage, Cap’n Andy Hawks is forced to dismiss the Bakers along with Pete, ending their time aboard the Cotton Blossom.

Magnolia Hawks, Julie’s best friend, rises to become the show’s new leading lady, and riverboat gambler Gaylord Ravenal is brought on as her partner. The two quickly find success, fall in love, and marry, leaving the boat to start a life in Chicago with Ravenal’s winnings. Their happiness is fragile, however, as Ravenal’s gambling habits eventually drain their finances, and he deserts Magnolia when the money runs out.

Ellie Shipley, once part of the show’s dance team, and her partner Frank Schultz, take Magnolia to audition at the Trocadero nightclub, inadvertently foreshadowing the arrival of Julie La Verne as a rival performer. Julie overhears Magnolia’s audition and learns from the nightclub manager that Ravenal has deserted her. In a calculated move, Julie quits so that Ravenal will be compelled to hire Magnolia again. Cap’n Andy Hawks attends Magnolia’s opening night, lending her support and helping her battle stage fright. Magnolia reveals she is pregnant with Ravenal’s child, and she returns to the show boat with Cap’n Andy, giving birth to a daughter named Kim.

Several years pass. Ravenal is back on a packet boat, and a drunken Julie struggles to sing on deck. After he punches an escort who slaps Julie, Ravenal goes out to the vessel’s deck, and Julie, who has been following Magnolia’s life, finally learns Ravenal’s true identity. She warns him, not realizing he knows nothing about Magnolia’s pregnancy, and his guilt drives him to reconnect. The next day he returns to Magnolia, and the couple reconciles as they face the future together, now with a family and a renewed sense of commitment.
