Scooby-Doo and the Mystery Inc. gang join forces with the iconic rock band KISS in an extraordinary adventure. At Kiss World, the Mystery Inc. gang—Frank Welker as Fred, Grey DeLisle as Daphne, Mindy Cohn as Velma, Matthew Lillard as Shaggy, and Frank Welker as Scooby-Doo—head to a sprawling amusement park built around the legendary rock group Kiss to witness a big Halloween concert and, more importantly, to crack a mystery. Daphne’s crush on Starchild, Paul Stanley, adds a playful tension for the team and for Fred, who would rather chase clues than chase romance.
Security chief Delilah Domino, Pauley Perrette, initially blocks their entry, citing safety concerns, and the gang sneaks inside anyway only to be caught and banned for supposed vigilantism. When Kiss themselves arrive, the heroes explain their purpose and Delilah reluctantly agrees to let them stay, though she remains wary of amateur detectives who might complicate her plans.
The Demon, [Gene Simmons], is wary of the newcomers after a peculiar earlier encounter when Scooby sprayed him with a water gun, but he later softens and agrees to a begrudging cooperation. As the group splits up for clues, Shaggy and Scooby are chased by a mysterious entity; Kiss rushes in to help, explaining that the terror is the Crimson Witch who has haunted Kiss World for some time and must disappear before the concert can proceed. Delilah, meanwhile, blames park supervisor Manny Goldman, [Garry Marshall], for the damage and threatens to shut down the park for safety—while vowing to track the gang down and arrest them.
A barefoot fortune-teller named Chikara, [Jennifer Carpenter], warns that the witch is from an alternate universe called Kissteria and plans to use the Black Diamond Kiss uses in their hit Detroit Rock City to summon a monster called The Destroyer to conquer Earth, a claim Velma struggles to accept. To stop the Crimson Witch, the gang uses the diamond to lure the witch, but she pursues them through a portal to Kissteria. There she steals the diamond and unleashes The Destroyer, sending the team and Kiss into a pulse-pounding spaceflight aboard a spaceship to halt the monster. They finally return to Earth, waking up as if the whole adventure might have been a gas-induced hallucination.
Back home, the gang unmasks the Crimson Witch as Delilah Domino, revealing that her true goal was to sell the Black Diamond to a competing defense company and to keep Mystery Inc. out of the loop while disguising her treachery as security concerns. Defeated, Delilah vows to sue Mystery Inc. for exposure, while Kiss agrees to handle the legal fallout in court.
In the final moments, Starchild shares a kiss with Daphne, and Daphne returns the affection with a kiss on Fred’s cheek, much to his delight. The Demon grins at Shaggy and Scooby, and Kiss sails off with the Black Diamond toward the Smithsonian. As they watch the legends depart, Shaggy turns to Scooby and asks whether they should tell Velma what they saw, but Scooby shrugs and smiles, “Why rock her world?”
