Upon returning to the museum, Larry discovers that many exhibits are being relocated to the Smithsonian Institution in Washington D.C. This change leaves behind the tablet of Ahkmenrah, which was key to the museum's magical nighttime displays, leaving Larry's animated friends vulnerable.

Larry receives a frantic call from Jedediah, who informs him that Dexter the monkey accidentally transported the tablet to the Smithsonian. The urgency escalates as they are under threat from Kahmunrah, Ahkmenrah's evil brother, who is intent on seizing the tablet's power.

Determined to help his friends, Larry dons the night guard uniform and sneaks into the Smithsonian archives. Upon arrival, he finds his beloved exhibits ensnared by Kahmunrah, who has plans for world domination using the tablet's magic.

Larry receives help from Major General George A. Custer and the adventurous Amelia Earhart. Together, they navigate the maze-like corridors of the Smithsonian, dodging Kahmunrah's soldiers and cleverly escaping through iconic historical photographs.

After a series of dramatic encounters, Kahmunrah captures Larry and presents him with a grim ultimatum: solve the tablet's combination in one hour or face Jedediah's dire fate. This tense moment is compounded by Amelia's confession of her feelings for Larry.
