Gene Autry is a principled singing cowboy who quits his oil company job after discovering a scheme to profit from the land around Anita's orphanage. He travels to Mexico to uncover the truth and protect the children. With help from Valdez and Frog, he uses wit and song to expose Carruthers' fraud and win support from the community. His actions drive the plot as he orchestrates a plan to prove the existence of oil and save the orphanage.

Anita Loredo helps run the orphanage in Mexico with Padre Dominic and cares deeply for the children. She confronts Carruthers and the oil promoters over unpaid royalties and the danger to the orphanage's future. Her romance with Gene adds emotional stakes to the mission.

Padre Dominic oversees the Mexican orphanage and explains the land royalty arrangement that ties the orphanage's fate to the oil deal. He informs Gene about the scheme and offers guidance on how to approach the investigation. He supports Gene's plan to expose the fraud and protect the orphans.

Valdez is a Mexican bandit who, under Gene's influence, becomes a modern Robin Hood. He helps raise donations at the fundraising dance and assists in pursuing the criminals. In the climax, Valdez sacrifices his life to save the children and proves himself a true friend.

Carruthers is the crooked head of Alta Vista Oil Company who schemes to profit from a land that the orphans depend on. He orders the kidnapping of Blythe to silence a whistleblower and manipulates stock to cling to power. When the truth is revealed and oil is found, his plans unravel as Gene and the others strike back.

Frog Millhouse is Gene's loyal sidekick who provides humor and support during the pursuit. He helps chase the villains and keeps spirits up at critical moments.

Blythe is Alta Vista's lead engineer whose testimony confirms there is oil on the site. He is kidnapped by Carruthers' men but is rescued, providing crucial evidence to back Gene's plan. His information helps drive the quest to drill and prove the land's value.
