A man on the streets searches for a prostitute and threatens her with a razor blade, revealing his aggressive intent. Derek Bliss intervenes by pointing an unusual-looking gun at the man's temple, causing him to back away in fear. The prostitute thanks Derek, and he quickly kills her with stakes, revealing she was a vampire.

Derek drags the vampire prostitute to an abandoned car lot and watches as her body catches fire in the sunlight. He films the scene, documenting the vampire's destruction to gather evidence of supernatural activity. This photo evidence is part of his ongoing effort to hunt vampires.

Back at his hotel, Derek receives a call from the Van Helsing Group leader informing him of a new mission. He prepares to head to Mexico, indicating he is a professional vampire hunter with ongoing assignments. The new job involves investigating vampire activity linked to a monastery.

Derek travels to a Mexican monastery where vampire-hunting priests are staying. That night, Una, a vampire princess, and her fledglings attack and feed on a man in the vicinity, demonstrating the ongoing threat posed by vampires and their violent nature.

Derek searches for people listed in a vampire-related dossier, but most are found dead or insane, and the last few are killed during his pursuit. His investigation reveals that the vampire threat is widespread and lethal, complicating his mission.

At a coffee shop, Derek meets Zoey, a young woman who is suspiciously familiar with the 'undead' business. He becomes suspicious of her and later discovers she's a vampire using a special lens. Zoey confesses she was bitten during a one-night stand, adding a new complication to the story.

Zoey flees after a confrontation, and shortly afterward, Una arrives and slashes the throats of all customers in the coffee shop, kidnapping Jesse, a man Derek was looking for. Zoe escapes during the chaos, hinting at her conflicted nature and her connection to the vampires.

Zoey shares her vision of a monastery, leading Derek to realize it is the one he just left. They return there and find all but one priest slaughtered. Father Rodrigo explains that Una is seeking the Béziers Cross, an artifact that could make vampires invincible by walking in daylight.

The group discovers a converted van filled with vampire-slaying tools, preparing for the confrontation. Derek meets Sancho, a teenage vampire hunter who joins their quest, and they team up with Ray Collins to hunt Una and her vampires.

Una seduces Ray Collins and manipulates him into giving her Zoey's pills, which suppress her vampire side. They reach the vampires' hideout, where Una captures Zoey. Derek is misled to believe Sancho gave the pills to Una, leading to a deadly confrontation.

Zoey's vampire blood is swapped with human blood in a clinic after her pills run out, temporarily suppressing her vampire side. Derek and the team work to save Rodrigo, a priest who is not truly one, from the vampires' trap, attempting to weaken Una's plans.

Derek fights to save Rodrigo from vampire attacks, culminating in a showdown with Una. She escapes into sunlight, dragging Derek into a final confrontation. Derek shoots her head off with a shotgun, causing her body to turn into stone with a beating black heart.

Derek kills Una by shooting her head into the sun, ending her threat. The others decide to stay in the village, while Derek and Zoey drive away at sunset to resupply on pills, emphasizing the ongoing danger and the need for constant vigilance.
