On a desolate road in New Mexico, a formidable demon disguised as a man, known as The Collector, relentlessly pursues drifter Frank Brayker. Their paths collide in a violent crash, prompting Brayker to make a hasty escape. He is aided by the local drunk, Uncle Willy, who brings him to a boarded-up church that has been repurposed into a boarding house. Here, he observes a ragtag group of residents, including the owner Irene, the sex worker Cordelia, the postal clerk Wally, and a convict named Jeryline who is out on work release.
A misogynistic cook called Roach barges in, announcing a theft attempt on his boss’s vehicle, blissfully unaware that it was Brayker trying to escape. Skeptical, Irene opts to contact the sheriff. Sheriff Tupper and his deputy Bob come across The Collector at the scene of the accident. The Collector cunningly convinces them that Brayker is a dangerous criminal, leading to an arrest. However, the situation escalates dramatically when The Collector murders Tupper by brutally piercing his skull.
Driven by the power of the key-like artifact in Brayker’s possession, The Collector spills his own blood in the sand, summoning a horde of demonic entities. Acting quickly, Brayker utilizes blood from the artifact to create a protective barrier around the boarding house, urging the residents to endure the terrifying night ahead. Unable to breach the defenses, The Collector resorts to using his psychic abilities to seduce and possess Cordelia, leading to savage consequences as she turns on her neighbors, killing Wally and gravely injuring Irene. Ultimately, Brayker is forced to end Cordelia’s life to protect the group.
As they flee through a network of old mine tunnels beneath the building, Jeryline discovers a frightened boy named Danny hiding there. However, the group is soon overwhelmed by other townsfolk who have fallen under demonic control, forcing them back into the church. With tension mounting, Brayker reluctantly divulges the backstory of the key artifact. He explains that after God created Earth, demons harnessed seven keys to channel cosmic power. Upon their discovery, God cast the demons out into the universe. The artifact Brayker holds is the final key needed to restore their dominion and was filled with the blood of Jesus Christ by a thief named Sirach. Throughout history, guardians of the key have replenished it with their blood when they die, and Brayker received it from his commanding officer during World War I.
As the horror unfolds, Danny disappears, prompting Jeryline to rally everyone. Meanwhile, Roach stealthily pilfers the key from Brayker’s bag. In the church attic, Irene and Bob stumble upon Wally’s plan to rob the post office, armed with an arsenal of weapons. Meanwhile, The Collector possesses Uncle Willy, who then attacks the group. In a desperate bid for survival, Roach makes a fateful deal with The Collector, trading his life for the key—but he is soon betrayed and killed.
The battle reaches a frenzied climax as Brayker reclaims the key amidst chaos. Irene and Bob heroically sacrifice themselves to fend off the remaining minions. In the attic, The Collector ensnares Danny, mortally wounding Brayker in the process, only for Jeryline to retaliate and kill him. As Brayker succumbs to his injuries, he initiates Jeryline as the new guardian of the key, rendering all blood seals inactive. Yet, The Collector overpowers her, claiming the key for himself. When he offers Jeryline a position at his side, she confronts him fiercely, spitting blood from the key into his face, which forces him to revert to his demonic form before being vanquished.
With dawn breaking, Jeryline rejuvenates the key with Brayker’s blood and boards a bus with her cat, locking the door behind her. Just down the road, the bus encounters a mysterious stranger (portrayed by Mark David Kennerly) who, looking strikingly similar to his predecessor and carrying an identical suitcase, declines the offer to board, claiming he’ll wait for the next one. A chilling realization washes over Jeryline as she recognizes him as the next Collector. Their eyes meet briefly before he begins to follow her, whistling the haunting theme song from the Tales from the Crypt television series.
